Well the solution has been found. As-per cypherpunks' instructions in the ticket, right-click on "torbrowser-install-5.0.1_en-US.exe", left click on Properties, then click on the Unblock button. Lastly, re-run as Administrator.
**Shakes fist at Vista!**

I've opened a ticket with bugtracker on torproject.org (bug #16847).
In the meantime, I've noticed the Portable version of Torbrowser installs just fine.
Google: torbrowser portable
The first result is from sourceforge, download the latest version from them (currently it's: 5.0.1). T

I too am on Vista (32-bit, Business Edition) and cannot start the setup program. I've also tried to run it as Administrator, even tried with compatibility mode, no luck.
I've also downloaded the 5.5alpha1, it too does not start.
Seems Vista is being left out?
